چکیده

Human carbonic anhydrase C and bovine carbonic anhydrase B were modified with the affinity label, bromoacetazolamide, and human carbonic anhydrase B with N-bromoacetylacetazolamide. Tryptic peptides from the modified enzymes, containing alkylated histidines, have been isolated by ion exchange and two-dimensional high voltage electrophoresispaper chromatography. The partial amino acid sequence of the peptide from alkylated bovine enzyme B has been found to be Met-Val-Asn-Asn-Gly-His-Ser-Phe-Asn-Val-Glu-TyrAsx-Asx(Glx,Asx,Ser)Lys-. In all probability the histidine in this sequence that reacted is His-64. Comparison of the amino acid composition of the tryptic peptides from the two human isoenzymes with the proposed sequences of these enzymes has made it possible to identify histidine-64 in human enzyme C as that reacting with bromoacetazolamide and histidine-67 in human enzyme B as that reacting with N-bromoacetylacetazolamide.

Irreversible inactivation of bovine carbonic anhydrase B by bromoacetazolamide.

Bromoacetazolamide effects rather quickly a partial, and more slowly an almost complete, irreversible inactivation of bovine carbonic anhydrase B (EC 4.2.1.1). Under identical conditions, the enzyme is not inactivated by bromoacetic acid or iodoacetamide, nor does it react significantly with these compounds. The zinc-free enzyme does not undergo irreversible binding with W-bromoacetazolamide an...

Covalent labeling of the active site of human carbonic anhydrase B with N-bromoacetylacetazolamide.

Two or possibly three equivalents of N-bromoacetylacetazolamide interact with two histidine residues located in the active site region of human carbonic anhydrase B. The most rapid reaction occurs at the l-nitrogen of a histidine residue and this is followed by a slower alkylation of the 3-nitrogen of the same histidine and the 3-nitrogen of a second histidine. From a reaction mixture of N-brom...

Carbon dioxide hydration activity of carbonic anhydrase: kinetics of alkylated anhydrases B and C from humans (metalloenzymes-isoenzymes-active sites-mechanism).

A stop-flow kinetic study was performed on the carbon dioxide hydration activity of the human carbonic anhydrase B isoenzyme carboxymethylated at its histidine(200), and of the human C isoenzyme carboxyketoethylated at its histidine(63). The Michaelis-Menten parameters determined between pH 5.6 and 8.7 showed striking differences between the native and the alkylated enzymes, as well as between ...

Reduced Erythrocyte Carbonic Anhydrase Activity by Swietenia Macrophylla Seeds in Diabetic Rats

Carbonic anhydrases (CAs, EC 4) are zinc containing metalloenzymes. The active site of these enzymes contains a Zn (II) ion Co-ordinated by three histidine residues and a water molecule/hydroxide ion. Four distinct gene families encode them and distributed throughout the phylogenetic tree. Presently 16 different CA isoforms of alpha family have been isolated and characterized in mammals. They c...

pH Dependence Study of the Kinetic Reaction of Bovine Carbonic Anhydrase with 2,2'-Dithiobispyridine in the Absence and Presence of Surfactants

The pH dependence study reveals that the Cys 206 sulphydryl group of bovine carbonicanhydrase in the native form is not exposed. During the reaction of 2,2'-dithiobispyridine (2-DTP) with the enzyme, there was no absorbance change recorded. In the presence ofsurfactants, the pH dependence profiles of the apparent second order rate constants, kapp, forthe reaction of 2-DTP with bovine carbonic a...
